No feeder cells and xeno-free culture medium
StemFit® medium was developed to produce a reliable and well-defined growth condition for human stem cells. It has all of the necessary components for the culture of embryonic stem (ES) cells as well as induced pluripotent stem (iPS) cells, and contains only xeno-free components—nothing from other animal sources. StemFit also eliminates the need for feeder cells. These important benefits lead to a reduction of variation in growth, and reduces concerns for contamination in the cultivation of stem cells. Less media volume needed
Due to the high-quality components and the ideal concentration of nutrients, the volume of StemFit required per plate is lower than that of conventional media, and even lower than that of other feeder-free media. A further 50% reduction in media consumption is the direct result of far fewer feeding steps during the week. This means that you use only 4.5 mL of StemFit versus 12 mL of conventional media, yielding a savings in reagents, time, and money.
Reproducible growth rate
The cultivation of stem cells using StemFit results in a very reproducible growth rate (below left), and consistency means perfect planning for your experiments. No more variation due to feeder cells! You’re better off without them; analyzing the morphology of stem cells cultivated in StemFit shows that colony shape and size are very similar to that of colonies grown on feeder cells (below right).
Free up your weekends
The maintenance of stem cells is very complicated and labor-intensive. Worst of all is the tedium of a feeding step during the weekends! StemFit allows a weekend devoid of stem cell media changes. Easy transition from feeder cells
The majority of the stem cells are still maintained in feeder-cell dependent cultivation. The transition from feeder-cell systems to non-feeder-cell systems is a snap with StemFit. Just switch the medium two days before passaging the cells and continue cultivating them in StemFit. After two to three passages the feeder cells will be completely diluted out and a pure stem cell culture will be obtained. No extra steps. No fuss. Just a few days of your normal protocol, and an increase in lab efficiency.
Compatible with many matrices
StemFit has been tested on many different matrices. As can be seen on the right, the fold expansion rate of cells cultured in StemFit is much higher when compared to Medium A from the market leading competitor. Different cell matrices were used as growing matrix and combined with StemFit or with Medium A. The fold expansion of 201B7 hiPS cells was determined after 7 days.
Less production of lactate and more consistent gene expression profile
The typical methods used for the cultivation of stem-cells provide plenty of opportunities for these cells to undergo stress. Every passage and growth period may introduce unwanted changes in gene expression profiles. The production of lactate, in particular, is the result of hypoxic stress. This often causes changes in the genome expression profile–or, worse still, leads to unwanted differentiation of the stem cells. The CGI Catapult Institute in London showed that there is considerably less lactate production when the cells are grown in StemFit.
They extended their studies to investigate the changes in gene expression caused by StemFit as compared to 4 commercially available media. Their results definitively showed that gene expression profile using StemFit was far more consistent. Less stress for your stem cells means less stress for you!
